JPH2 encodes junctophilin-2, a membrane tethering protein that stabilizes junctional membrane complexes between the plasma membrane/T-tubules and sarcoplasmic reticulum in striated muscle. By maintaining nanoscale coupling between L-type Ca²⁺ channels and ryanodine receptors, JUNCTOPHILIN-2 supports excitation–contraction coupling and efficient intracellular Ca²⁺ release. JPH2 function intersects with Ca²⁺ handling pathways that influence contractility, electrical signaling, and downstream calcium-dependent transcriptional programs. Altered JPH2 expression or organization has been linked to cardiomyocyte T-tubule remodeling and dysregulated Ca²⁺ homeostasis, features associated with inherited and acquired cardiomyopathies and arrhythmogenic phenotypes in experimental models.
Junctophilin-2 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the JPH2 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the JPH2 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the JPH2 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Junctophilin-2 protein expression.
This CRISPR knockout system enables efficient generation of JPH2-deficient cell models for investigation of Junctophilin-2 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
