IscU1/2 Antibody (B-12) is a mouse monoclonal IgG1 kappa light chain antibody that detects IscU1/2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). anti-IscU1/2 antibody (B-12) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. IscU1 and IscU2 proteins play a crucial role in the assembly of iron-sulfur (Fe-S) clusters, which are essential cofactors involved in numerous biological processes, such as electron transfer in oxidative phosphorylation and enzymatic reactions in key metabolic pathways. IscU1 and IscU2 proteins are primarily localized in the mitochondria, where these proteins facilitate the assembly of Fe-S clusters by interacting with IscS, a cysteine desulfurase that provides inorganic sulfur. Proper functioning of IscU1 and IscU2 is vital, as deletion leads to an accumulation of iron in mitochondria, loss of aconitase activity, and increased oxidative stress in cells lacking cytosolic copper/zinc superoxide dismutase. In humans, alternative splicing of IscU pre-mRNA generates two distinct proteins that localize to different cellular compartments, with IscU1 found in the cytosol and IscU2 in the mitochondria. This differential localization underscores the importance of IscU proteins in maintaining cellular iron homeostasis and protecting against oxidative damage, highlighting their significance in cellular health and disease.
