IL-1α Antibody (ALF-161) is a hamster monoclonal IgG antibody that detects IL-1α of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), flow cytometry (FCM), and enzyme-linked immunosorbent assay (ELISA). IL-1α Antibody (ALF-161) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. IL-1α plays a pivotal role in the immune response and inflammation, acting as a key mediator that influences the activity of T and B lymphocytes as well as natural killer (NK) cells. IL-1α is primarily located in the cytoplasm of cells and can be released upon cell activation, which is crucial for mediating inflammatory responses. IL-1α presence in the cytoplasm allows for rapid release during inflammatory stimuli, enabling swift communication between immune cells and the surrounding tissue. IL-1α is involved in various physiological processes, including kidney development and cartilage degradation, and has been implicated in several pathological conditions such as rheumatoid arthritis, inflammatory bowel disease, and atherosclerosis. IL-1α binding to the same receptor as IL-1β, despite their distinct genetic origins, underscores IL-1α importance in orchestrating a coordinated immune response.

Can IL-1α (ALF-161): sc-12741 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, IL-1α (ALF-161): sc-12741 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.

Rated 5 out of 5 by from Mutant BRAF and MEK Inhibitors Regulate the TumorProtein lysates were prepared in Laemmli sample buffer, separated by SDS-PAGE, and proteins were transferred to polyvinylidene dif-luoride membranes. Immunoreactivity was detected using HRP-con-jugated secondary antibodies (CalBioTech) and chemiluminescence substrate (Thermo Fisher Scientific) on a Versadoc Imaging Sys-tem (Bio-Rad). Primary antibodies, all from Cell Signaling Technol-ogy unless otherwise stated, were as follows: anti-phospho-ERK1/2 (T202/Y204), anti-ERK2 (Santa Cruz Biotechnology), anti-GSDME (Abcam), anti–cleaved caspase-3, anti-BIM/BOD (Enzo Life Sci-ences), anti-IL1α (Santa Cruz Biotechnology), anti-HMGB1, and anti-GAPDH. Cell supernatants were harvested in the absence of FBS in culture medium to avoid distortion of SDS-PAGE. After centrifu-gation to remove cell debris, cell supernatants were concentrated 10×using Amicon Ultra 10K (Sigma-Aldrich). Concentrates were mixed with Laemmli sample buffer (Bio-Rad) and analyzed via Western blotting. For T-cell culture, concentrates were further washed twice with PBS and once with RPMI-1640 medium by spin, and sterilized by filtration through a 0.2-μm filter. Protein gel staining was per-formed using Coomassie Brilliant Blue R-250.
