IL-12B p40 Antibody (F-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects IL-12B p40 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). IL-12B p40 (F-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The interleukin-12 (IL-12) family, which includes IL-12B p40, plays a crucial role in the immune response, particularly in the differentiation of naive CD4(+) T cells into type 1 helper T cells that produce interferon-γ (IFN-γ). This process is vital for the immune system′s ability to combat intracellular pathogens and tumors. IL-12B p40 subunit is not only a component of IL-12 but also combines with p19 subunit to form IL-23, a cytokine that shares some functional properties with IL-12, such as the activation of transcription factor Stat4 and promotion of IFN-γ production. IL-12B p40′s ability to form these biologically active complexes underscores its importance in regulating immune responses and highlights anti-IL-12B p40 antibody (F-10)′s relevance in research focused on immunology and therapeutic interventions.

Here's question that my western result of mIL-12 in the cell lysate extracted from mouse lungs always shows about 50 kDa different from that of datasheet. Do you have any recommendation? Thank you!!

Thank you for your question. We have not tested our mouse monoclonal IL-12B p40 Antibody (F-10): sc-365389 in mouse lung tissue extracts. Biologically active IL-12 is a heterodimer composed of two covalently linked proteins; a 35-kDa light chain (known as p35 or IL-12 ) and a 40-kDa heavy chain (known as p40 or IL-12 ). Under reducing conditions, the expected molecular weight detected by this IL-12B p40 antibody is ~ 40 kDa. We recommend using the primary antibody at a dilution of 1:200, and loading 50 µg of protein. A link to our complete recommended protocol for Western blotting with this antibody is below. What is the recommended fixation method for IF with IL-12B p40 (F-10): sc-365389 monoclonal antibody?
