Each CRISPR/Cas9 Knockout (KO) Plasmid product consists of a pool of 3 plasmids designed to ensure identification and cleavage of a specific gene for maximum knockout efficiency. Each plasmid encodes a unique 20 nt sequence derived from the GeCKO (v2) library.
IGF-1 Receptor α/β/IGF1R CRISPR Plasmids (h)

- CRISPR/Cas9 KO Plasmids consists of IGF-1 Receptor α/β/IGF1R-specific 20 nt guide RNA sequences derived from the GeCKO (v2) library
- For CRISPR gene knockout, gRNA sequences direct the Cas9 protein to induce a site-specific double strand break (DSB) in the genomic DNA

- IGF-1 Receptor α/β/IGF1R CRISPR/Cas9 Knockout (KO) Plasmid (h2) consists of a pool of three plasmids each encoding the Cas9 nuclease and a IGF-1 Receptor α/β/IGF1R-specific 20 nt guide RNA (gRNA) designed for maximum knockout efficiency
- gRNA sequences are derived from the GeCKO (v2) library and direct the Cas9 protein to induce a site-specific double strand break (DSB) in the genomic DNA
- For selection of cells containing a successful Cas9-induced DSB, co-transfection with IGF-1 Receptor α/β/IGF1R HDR Plasmid (h2) (sc-400084-HDR-2) is recommended
- Following transfection, gene knockout efficiency can be assayed by WB, IF or IHC using antibody: IGF-1 Receptor/IGF1R Antibody (7G11): sc-81464

- Each HDR Plasmid inserts a puromycin resistance gene to enable selection of stable knockout (KO) cells and an RFP (Red Fluorescent Protein) gene to visually verify transfection
