HPI-1 is a Hedgehog pathway inhibitor Gli1/Gli2 inhibitor. This compound is a potent inhibitor of hedgehog signaling, effectively blocking the activation of the Sonic hedgehog (Shh), SAG, Gli2, and Gli1 pathways with IC50 values of 1.5 μM, 1.5 μM, 4 μM, and 6 μM respectively. Additionally, this inhibitor effectively suppresses Hh pathway activation induced by Shh, SAG, Gli2, and Gli1 in Shh-LIGHT2 cells, with IC50 values of 1.5 μM, 1.5 μM, 4 μM, and 6 μM respectively. It also inhibits Hh pathway activation in SmoM2-LIGHT cells, with an IC50 of 2.5 μM.
