SEE ALSO...

HLA-DMα Antibody (NB-B33) is a mouse monoclonal IgG1 kappa light chain antibody that detects the HLA-DM alpha protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Anti-HLA-DM alpha antibody (NB-B33) is available as a non-conjugated format. HLA-DM alpha protein plays a crucial role in the immune response by facilitating the loading of peptides onto major histocompatibility complex (MHC) class II molecules, which are essential for the activation of CD4+ T helper cells. This process is vital for the adaptive immune system, as HLA-DM alpha enables the presentation of extracellular antigens to T cells, thereby initiating a targeted immune response. HLA-DM alpha protein is primarily located in endosomal and lysosomal compartments, as well as on the surface of B cells, where HLA-DM alpha interacts with MHC class II molecules to promote the exchange of peptides. Proper functioning of HLA-DM alpha protein is critical for maintaining immune tolerance and preventing autoimmune diseases, as evidenced by studies linking specific polymorphisms in HLA-DM genes to susceptibility to conditions such as rheumatoid arthritis. Using HLA-DMα (NB-B33) monoclonal antibody in research can provide valuable insights into the mechanisms of antigen presentation and the role of HLA-DM alpha in immune regulation.

Can sc-134356: HLA-DMα (NB-B33) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: jerojero
Thank you for your question. Yes, sc-134356: HLA-DMα (NB-B33) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
