    HHV-8 K14 Antibody (OX113) is a mouse monoclonal IgG1 antibody that detects HHV-8 K14 protein of viral origin by immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). HHV-8 K14 (OX113) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. HHV-8, also known as Kaposi′s sarcoma-associated herpesvirus, is linked to several serious conditions, including multicentric Castleman′s disease and primary effusion lymphoma, a rare form of non-Hodgkin lymphoma that affects body cavities. HHV-8 K14 protein is notably expressed on the surfaces of infected cells exclusively during the lytic cycle, which is crucial for viral replication and spread. HHV-8 K14 interacts with human CD200R, a receptor found on myeloid cells that plays a significant role in modulating immune responses by locally restricting macrophage activation. The structural similarity between HHV-8 K14 and CD200, which shares 40% sequence identity, allows HHV-8 to effectively mimic CD200 activity, thereby inhibiting tumor necrosis factor-alpha production and dampening the immune response. This interaction is vital for viral evasion of the host immune system, highlighting HHV-8 K14′s importance in viral pathogenesis and potential as a target for therapeutic intervention.

    The immunostain must be nuclear or may be cytoplasmic ???

    Thank you for your question. The expected localization when using this antibody, HHV-8 K14 (OX113): sc-53072, is diffuse cytoplasmic staining. (Refer to PMID: 18077714, Table 1).

    What negative control should be used when using the conjugated version of HHV-8 K14 (OX113): sc-53072 monoclonal antibody in flow cytometry?

    Thank you for your inquiry. We recommend using the isotype specific normal mouse IgG conjugated to the same fluorophore. For example, if you are using sc-53072 PE, then you would want to use normal mouse IgG1 PE, sc-2866.

    Can HHV-8 K14 (OX113): sc-53072 this antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

    Thank you for your question. Yes, HHV-8 K14 (OX113): sc-53072 monoclonal antibody has been validated for use in IHC with paraffin-embedded sections.
