Gum guaiac is a tree resin and crude source of 2,5-di-(4-hydroxy-3-methoxyphenyl)-3,4-dimethylfuran (α-guaiaconic acid). It produces the phenolic α-guaiaconic acid, which acts as a colorimetric peroxidase substrate. The α-guaiaconic acid reacts with organic or inorganic oxidizing agents by oxidizing to a blue-colored quinone. Gum guaiac can begin a reaction that detects trace amounts of heme from hemoglobin in the presence of peroxide. Gum guaiac (67% w/v ratio) diluted in 96% ethanol can perform the Nobles test to analyze extracellular oxidase production.
