GRAP1 Antibody (A-6) is a mouse monoclonal IgG2a kappa light chain antibody that detects GRAP1 of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). GRAP1 (A-6) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. GRAP1, also known as GRASP-1 or GRIP-associated protein, plays a crucial role in cellular signaling as a member of the rasGEF (Ras protein GDP/GTP exchange factors) family, which is essential for the regulation of Ras signaling pathways. GRAP1 contains a unique structural arrangement featuring a rasGEF domain, a caspase-3 cleavage site, a region homologous to the ras binding domain, and a PDZ domain. This structural configuration is significant because the caspase-3 cleavage site divides GRAP1 into an amino-terminal GEF catalytic domain and a carboxyl-terminal regulatory domain, which is a proteolytic fragment. The presence of these domains allows GRAP1 to interact with other proteins, such as the seventh PDZ domain of GRIP-1, which binds to the C termini of AMPA receptors, potentially linking them to other signaling molecules. GRAP1 is predominantly expressed in the cytosol and is partially localized to the membrane in all tissues of the nervous system, while the cleaved fragment remains in the cytosol. The regulation of GRAP1 by caspase-3 is particularly important, as this process may disrupt the proper targeting and function of the GEF, thereby influencing the distribution of AMPA receptors and the overall signaling dynamics within neuronal cells.
