granulysin Antibody (F-9) is a mouse monoclonal IgG2a kappa light chain antibody that detects granulysin protein of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). granulysin Antibody (F-9) is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. granulysin, also known as NKG5, is a saposin-like protein (SAPLIP) predominantly expressed in cytotoxic lymphocytes, where granulysin plays a crucial role in immune response against intracellular pathogens. granulysin mediates the release of cytotoxic granules from cytolytic T lymphocytes (CTLs) and natural killer (NK) cells upon stimulation by antigens from bacteria, fungi, and parasites. granulysin′s mechanism of action involves increasing the permeability of pathogen host cells, leading to their lysis and subsequent apoptosis. granulysin is produced in two distinct sizes from three unique transcripts, highlighting complex regulation. granulysin-expressing CTLs, in conjunction with perforin, can effectively target and eliminate both extracellular and intracellular Mycobacterium tuberculosis, underscoring granulysin′s significant role in T cell-mediated immunity and host protection against various pathogens.

Does this antibody require any sort of HIER or enzyme epitope retrieval? Thank you!

Thank you for your question. Yes, we do recommend antigen retrieval. Heat treatment (recommended method): Place slides in a container and cover with 10 mM sodium citrate buffer, pH 6.0 (sc-294091); or with 50 mM glycine-HCl buffer (sc-29096), pH 3.5, with 0.01% (w/v) EDTA (sc-29092). Heat at 95° C for 5 minutes. Top off with fresh buffer and heat at 95° C for 5 minutes (optimal incubation time may vary for each tissue type). Allow slides to cool in the buffer for approximately 20 minutes. Wash in deionized H2O three times for 2 minutes each. Aspirate excess liquid from slides.

I am using granulysin (F-9): sc-271119 monoclonal antibody to stain paraffin embedded tissue. Do you recommend antigen retrieval when using this antibody?
