Granisetron is a serotonin 5-HT3 receptor antagonist that functions by selectively blocking the action of serotonin at 5-HT3 receptors. This prevents the initiation of a signal cascade that would otherwise lead to nausea and vomiting. By binding to the receptor, granisetron inhibits the activation of the vagus nerve, which plays a key role in the emetic reflex. At the molecular level, granisetron competes with serotonin for binding to the 5-HT3 receptor, thereby preventing the downstream signaling events that would normally lead to emesis.
