GP49 Antibody (H1.1) is a hamster monoclonal IgG antibody that detects GP49 protein of mouse origin by flow cytometry (FCM). GP49 (H1.1) antibody is available in non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates, providing versatility for different experimental needs. GP49 protein is a type I transmembrane glycoprotein belonging to the immunoglobulin superfamily, primarily expressed on myeloid cells such as mast cells, natural killer (NK) cells, and macrophages, which play crucial roles in both innate and adaptive immunity. GP49 exists in two major isoforms, GP49A and GP49B, encoded by distinct genes that exhibit approximately 95% homology. GP49B serves as an inhibitory isoform, characterized by two immunoreceptor tyrosine-based inhibitory motifs (ITIMs) in its C-terminal region, which are essential for dampening immune responses. GP49A lacks these inhibitory motifs and has a shorter cytoplasmic domain, allowing potential homodimer formation that can trigger calcium mobilization, eicosanoid production, and cytokine gene transcription, thereby influencing immune signaling pathways. GP49B is homologous to the human Fc receptor for IgA, HM18, which underscores GP49′s importance in immune regulation and potential therapeutic applications.

How can GP49 (H1.1): sc-53584 mouse monoclonal antibody be used for double or triple staining, if the other primary antibodies are also raised in mouse?

In this case, we recommend using a directly conjugated mouse monoclonal primary antibody. This antibody is available conjugated to phycoerythrin (sc-53584 PE) or fluoroscein (sc-53584 FITC).
