GM 1489 is a potent broad-range inhibitor of MMP (matrix metalloproteinases). It inhibits MMPs in vitro (Ki = 0.2 nM for MMP-1 Ki = 500 nM for MMP-2 Ki = 20 muM for MMP-3 Ki = 100 nM for MMP-8 and Ki = 100 nM for MMP-9). Additionally, it reduces 5-aza-2′-deoxycytidine-induced increases in MMP-1, MMP-2, MMP-3, MMP-7, MMP-9, and MMP-14 expression as well as cell invasion in AsPC-1, BxPC-3, Hs766T, MiaPaCa2, and PANC-1 cancer cells. GM 1489 also reduces the inflammatory and the hyperproliferative responses that occur following topical phorbol ester application.
