Gly-Phe-β-naphthylamide is a cathepsin C substrate that is described to accumulate inside lysosomes and cause their destruction. Cathepsin C cleaves the dipeptide bond of Gly-Phe-β-naphthylamide, releasing 2-naphthylamine. The release of 2-naphthylamine can be monitored spectrophotometrically, allowing for the measurement of cathepsin C activity. It has also been used to study recombinant cathepsin C. Studies report that Gly-Phe β-naphthylamide significantly inhibits the activation of caspase-8.
