Gly-Phe-AFC, a cell-permeant fluorogenic peptide substrate, has been developed for the quantification of viable cells in cytotoxic assays. Upon entering intact cells, this substrate undergoes cleavage, leading to the production of a fluorescent signal that is directly proportional to the number of living cells. Serving as a reliable marker of live-cell protease activity, it loses functionality when membrane integrity is compromised, preventing leakage into the surrounding culture medium. Additionally, Gly-Phe-AFC serves as a substrate for measuring the activity of Dipeptidyl peptidase 1 (DPP1), an intracellular lysosomal cysteinyl protease from the papain family involved in the degradation of proteins.
