Datasheets
  • Glucosidase I Antibody (H-6) is a mouse monoclonal IgG1 κ provided at 200 µg/ml
  • raised against amino acids 395-694 mapping within an internal region of Glucosidase I of human origin
  • recommended for detection of Glucosidase I of mouse, rat and human origin by WB, IP, IF, IHC(P) and ELISA
  • m-IgG Fc BP-HRP and m-IgG1 BP-HRP are the preferred secondary detection reagents for Glucosidase I Antibody (H-6) for WB and IHC(P) applications. These reagents are now offered in bundles with Glucosidase I Antibody (H-6) (see ordering information below).

    SEE ALSO...

Glucosidase I Antibody (H-6) is a mouse monoclonal IgG1 antibody that detects Glucosidase I in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Glucosidase I plays a crucial role in the N-linked glycosylation pathway, specifically catalyzing the removal of the distal alpha 1,2-linked glucose residue from the Glc3-Man9-GlcNAc2 oligosaccharide precursor within the lumen of the endoplasmic reticulum. This enzymatic activity is vital for proper protein folding and maturation, ensuring glycoproteins acquire the correct oligosaccharide structures necessary for stability and function. Disruptions in Glucosidase I activity due to genetic mutations can lead to congenital disorders of glycosylation, such as CDG-IIb, which manifest in severe clinical symptoms including hypotonia, dysmorphic features, and life-threatening complications. The structural integrity of Glucosidase I, characterized by a short cytosolic tail, single-pass transmembrane domain, and large C-terminal catalytic domain, is essential for function, as alterations can significantly impact glycoprotein processing. Glucosidase I (H-6) antibody serves as an invaluable tool for researchers studying glycosylation processes and related disorders.

Hello, i work with S. pombe and T. cruzi as model organisms. I need a GI antibody for both western blot and immunofluorescence. I ran structural alignments and despite not having aminoacidic identity (25%), structural is 80%. Would this Ab work for me?

    Asked by: Tommy
Thank you for your question. sc-365399 Glucosidase I (H-6) is recommended for western blot, IP and ELISA in mouse, rat and human samples .

Hello, i bought this antibody a couple of months ago and i was wondering what amount of total protein do you use from the cells lysate to detect presence of GI. I tried with 30ug and couldn't get signal.

    Asked by: Tommy
Thank you for your question. We use the following amounts in western blots, depending on sample type: 40–60 µg whole cell lysate, 10–20 µg nuclear extract, 5-10 µg transfected lysate, or 10–20 ng purified protein per lane.

Can Glucosidase I (H-6): sc-365399 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

    Asked by: Trav11
Thank you for your question. Yes, Glucosidase I (H-6): sc-365399 has been validated for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining
