Gigaxonin Antibody (F-3) is a mouse monoclonal IgG2b kappa light chain antibody that detects Gigaxonin of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). Gigaxonin (F-3) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Gigaxonin, also known as giant axonal neuropathy, GAN1, or KLHL16, plays a critical role in protein degradation regulation, which is essential for maintaining neuronal function and survival. Gigaxonin belongs to the cytoskeletal BTB/kelch repeat family and influences cytoskeletal organization and dynamics, particularly in neurofilament architecture. The amino-terminal BTB domain binds to ubiquitin-activating enzyme E1, while the carboxy-terminal kelch repeat domain interacts with microtubule-associated protein 1B (MAP1B) light chain, marking MAP1B for degradation. This interaction remains crucial because MAP1B overexpression can lead to neuronal cell death, whereas reduced MAP1B levels significantly enhance neuronal survival. Mutations in Gigaxonin cause human giant axonal neuropathy (GAN), an autosomal recessive neurodegenerative disorder characterized by axonal degeneration due to cytoskeletal abnormalities, including intermediate filament accumulation.

Can Gigaxonin (F-3): sc-376173 monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Thank you for your question. Yes, Gigaxonin (F-3): sc-376173 has been validated for use in IHC with paraffin-embedded sections. Rated 4 out of 5 by from Good clean antibodyTested a free sample on SH-SY5Y lysate and bacterial lysate of recombinant protein. The antibody detected a single band at the appropriate migration in the cell lysate and had a strong reaction with 50ng of fusion protein. Gigaxonin is not highly expressed, but this antibody worked better than expected at a 1:500 dilution.
