Growth arrest–specific 6 (GAS6) encodes Gas6, a vitamin K–dependent secreted ligand for the TAM receptor tyrosine kinases AXL, MERTK, and TYRO3. Gas6–TAM signaling regulates efferocytosis, innate immune dampening, cell survival, and migration through pathways that commonly include PI3K–AKT, MAPK/ERK, and NF-κB, with strong effects on macrophage polarization and endothelial homeostasis. In human tissues, GAS6 activity contributes to thromboinflammatory crosstalk and can reshape tumor–immune interactions and metastatic behavior via AXL/MERTK-dependent programs. Dysregulated GAS6/TAM axis signaling has been linked to cancer progression, chronic inflammation, fibrosis, and vascular pathobiology, making it a frequent target in mechanistic studies of microenvironmental signaling.
Gas6 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the GAS6 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the GAS6 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the GAS6 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Gas6 protein expression.
This CRISPR knockout system enables efficient generation of GAS6-deficient cell models for investigation of Gas6 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
