Fluorescein-O′-acetic acid (FOA) is a fluorescent label which reacts with amines after activation to give isomerically homogeneous derivatives. It is used to study the metabolism of cells, to identify and quantify cellular components, and to measure enzyme activities. FOA is taken up by cells and converted to fluorescein-O-acetyl-ester. This is then hydrolyzed by intracellular esterases to form fluorescein, which is excreted from the cell. This process results in the cell accumulating a fluorescent signal that can be used to study cellular processes. Fluorescein-O-acetyl-ester may act to inhibit the activity of some enzymes, increase the permeability of cell membranes, induce apoptosis in some cell types, and induce the production of reactive oxygen species in cells.
