FLUO 3/AM, a visible wavelength calcium probe, can be detected through fluorescent cytometry. Experiments show that this compound can be loaded into lymphocytes and fibroblasts by incubation with the pentaacetoxymethyl ester of the dye. Once inside the cell, the ester is hydrolyzed to yield genuine FLUO-3/AM capable of indicating changes in Ca2+. Other studies have reported that this FLUO 3/AM monitors Ca2+ levels without interfering with intracellular conditions. Additionally, this agent has been noted to be an effective probe for assessing the concentration and kinetics of change in mitochondrial Ca2+ under variety of conditions.
