FBPase Antibody (F-9) is a mouse monoclonal IgG2b antibody that detects fructose-1,6-bisphosphatase (FBPase) in human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). FBPase plays a crucial role in carbohydrate metabolism by catalyzing the conversion of fructose-1,6-bisphosphate into fructose 6-phosphate and inorganic phosphate, a key step in gluconeogenesis and glycolysis. This enzymatic activity is vital for maintaining glucose homeostasis, particularly during fasting or intense exercise, as FBPase helps regulate blood sugar levels. FBPase is encoded by two genes, FBP1 and FBP2, which produce liver and muscle isoforms, respectively, and is present in all living organisms. Notably, the liver isoform consists of four identical subunits, and AMP modulates FBPase activity by inhibiting the enzyme and affecting substrate turnover without altering substrate affinity. Mutations in the FBP1 gene can lead to an autosomal recessive disorder characterized by FBPase deficiency, resulting in serious metabolic complications such as hypoglycemia and metabolic acidosis. The muscle isoform of FBPase is strategically located on both sides of the Z-line, which is essential for muscle metabolism function.
