Estetrol is a metabolite of Estradiol that is an estrogenic steroid synthesized exclusively by the fetal liver during human pregnancy and reaches the maternal circulation through the placenta. It binds to the estrogen receptor and as a selective estrogen receptor modulator (SERM), it exhibits estrogen agonism in certain tissues and estrogen antagonism in others. Estetrol displays weak estrogen activity in the uterus and acts as an estrogen antagonist in breast tissue.
