EpoR Antibody (D-5) is a mouse monoclonal IgG3 kappa light chain antibody that detects EpoR of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). EpoR (D-5) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The erythropoietin receptor (EpoR) plays a crucial role in erythropoiesis, as EpoR mediates the effects of erythropoietin (Epo), a key hormone in red blood cell production. EpoR is a 507 amino acid transmembrane protein that undergoes homodimerization upon Epo binding, which is essential for initiating downstream signaling pathways that lead to cell proliferation and survival. Notably, EpoR shares significant structural similarities with the interleukin 2 receptor beta-chain subunit, particularly in the conserved box 1 and box 2 domains of their cytoplasmic regions, which are critical for receptor function. These conserved domains facilitate the coupling of ligand binding to the induction of tyrosine phosphorylation, a key step in signal transduction. Additionally, the membrane proximal region of EpoR is vital for the recruitment of Janus kinase 2 (JAK2), further underscoring the importance of EpoR′s structure in functional role. The presence of various cross-linked complexes and differing ligand affinities indicates that EpoR may operate as part of a multireceptor complex, highlighting EpoR′s intricate involvement in cellular signaling and the regulation of erythropoiesis.

datasheet 상에는 application 에 human 만 되어있는데, 다른 레퍼런스 확인해보니 mouse나 rat 에서 작동하더라구요. 저희 연구실에 마우스 조직을 이용한 western blot 진행 시 45 kda 사이즈에서 밴드가 나타나는데 human 과는 달라서 그런지 60kda 사이즈가 아니더군요. 다른 종에서도 작동하는게 맞나요?

Thank you for your question. sc-365662 is recommended for detection of EpoR of human origin by WB, IP, IF and ELISA.To determine whether it is the target band, strict positive and negative controls as well as knockout experiments are required for verification.

I used to use M-20 and C-20 to detect total EPOR in my IP samples, however, they are all discontinued, i tried this D-5 many times, but it doesn´t work at all. Do you have other recommendations? I use human cell line ASE2.

All of the other antibodies for rat EPO-R of your company are now discontinued. Does this antibody sc-365662 cross-react with rat EPO-R in western blot ?

Thank you for your question. We have not yet tested this antibody, EpoR (D-5): sc-365662, for reactivity with EpoR of mouse or rat origin. Therefore, it is not a recommended species at this time. Unfortunately, we do not offer any alternative antibodies for rat EpoR at this time.

Is this antibody good for flow cytometry?

This indicates there will be problems with false-positive results. SCBT should add a real positive control on the website to justify the conclusions.
