envoplakin Antibody (5G96) is a mouse monoclonal IgG1 antibody that detects envoplakin in human samples through applications such as western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Envoplakin is a crucial membrane-associated precursor of the epidermal cornified envelope and desmosomes, playing a significant role in maintaining the structural integrity of epithelial tissues. Envoplakin is predominantly expressed in epidermal and esophageal keratinocytes, as well as in nonepithelial stratified squamous epithelia, but is absent in simple epithelia and nonepithelial cells. Envoplakin colocalizes with desmoplakin at desmosomes and along keratin filaments throughout the differentiated layers of the epidermis, contributing to cell adhesion and tissue resilience. Notably, envoplakin forms nuclear and cytoplasmic aggregates with associated intermediate filaments, a process influenced by its rod domain, which is essential for aggregation, and its linker domain, which facilitates intermediate filament association. Furthermore, envoplakin distribution at the interdesmosomal plasma membrane depends on heterodimerization with periplakin, highlighting the intricate interplay between these proteins in maintaining cellular architecture.
