Eniluracil, a potent inhibitor of dihydropyrimidine dehydrogenase (DPD) with a Ki of 1.6 µM, plays a role in the breakdown of pyrimidines, such as 5-fluorouracil. This oral fluoropyrimidine analogue inhibits liver DPD, leading to increased plasma pyrimidine levels in rats, with ED50s of 1.8 and 10 µg/kg, respectively. The significant impact of eniluracil becomes evident as it enhances the antitumor effects of 5-fluorouracil in MC-38 murine colon carcinoma and MOPC 315 murine myeloma models. The primary function of eniluracil is to inactivate DPD, the rate-limiting enzyme responsible for the breakdown and inactivation of 5-fluorouracil (5-FU) in the liver.
