eIF6 Antibody (A-12) is a mouse monoclonal IgG1 antibody that detects eIF6 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). eIF6, also known as CAB, B2GCN homolog, p27(BBP), or B4 integrin interactor, is a crucial 245 amino acid protein primarily localized in the cytoplasm and nucleolus of the nucleus, where eIF6 plays a significant role in protein synthesis regulation. eIF6′s structure features distinct N-terminal and C-terminal subdomains harboring essential nucleolar localization sequences, vital for ribosomal assembly function. Through binding to the 60S ribosomal subunit, eIF6 facilitates translocation from the nucleus to cytoplasm while preventing premature association with the 40S subunit, thereby inhibiting 80S initiation complex formation. This regulation influences both ribosomal function and transcription processes. In the cytoplasm, the eIF6-60S complex undergoes phosphorylation via the RACK1/PKC pathway, leading to eIF6 dissociation from the ribosomal subunit, essential for translation initiation. eIF6 upregulation has been strongly linked to various cancers, including ovarian cancer, indicating eIF6′s potential role in carcinogenesis and significance as a cancer research target.
