E-64-c is an effective synthetic calpain inhibitor composed from E-64 (sc-201276), which also inhibits cathepsin B, cathepsin H, and cathepsin L (cysteine proteases). The compound has been demonstrated to form a complex with papain by binding via a thioether covalent bond. Studies indicate that E-64-c, also known as Ep-475, is a more potent inhibitor of cathepsins B and L compared to E-64. Mouse lymphosarcoma LS studies show that E-64-c decreases the effects of cyclophosphamide.
