DPH2 Antibody (6E7) is a mouse monoclonal IgG2a kappa light chain antibody that detects DPH2 protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), and immunofluorescence (IF). Anti-DPH2 antibody (6E7) is available as the non-conjugated monoclonal isotype antibody. DPH2, also known as diphthamide biosynthesis protein 2 or DPH2L2, is a 489 amino acid protein that plays a crucial role in the biosynthesis of diphthamide, a unique post-translational modification of histidine found in elongation factor 2 (EF-2). This modification is essential for proper function of EF-2, which is involved in protein synthesis, and serves as a target for diphtheria toxin, highlighting DPH2′s importance in cellular response to toxins. DPH2 exhibits strong expression in skeletal muscle and moderate expression in various tissues, including heart, small intestine, liver, pancreas, testis, and colon, while showing lesser expression in brain, placenta, kidney, spleen, thymus, prostate, ovary, and lymphocytes. DPH2 interacts with DPH1, and together they function as a dimer or multimer, participating in diphthamide biosynthesis pathway. Loss of DPH2 in Saccharomyces cerevisiae has been shown to suppress zymocicity, indicating potential role in cellular health and disease. Two transcript variants encoding different isoforms of DPH2 have been identified, further underscoring complexity and significance in biological processes.
