Donepezil Hydrochloride is a piperidine derivative and a noncompetitive AChE (acetylcholinesterase) inhibitor (IC50 = 11.6 nM). Studies indicate that donepezil hydrochloride selectively inhibits AChE (acetylcholinesterase), whereas it has little effect on butyrylcholinesterase. Donepezil binds to hydrophobic sites on the AChE enzyme without altering the structure of either the enzyme or the inhibitor, allowing for a dynamic process of inhibition This mechanism of action prevents the breakdown of acetylcholine. Alternate studies suggest that donepezil hydrochloride increases the concentration of extracellular acetylcholine. Acetylcholine is a neurotransmitter involved in memory and learning processes. Donepezil Hydrochloride′s metabolic process involves cytochrome P450 enzymes.
