Human DMTF1 encodes Dmp1, a Myb-like transcription factor that regulates cell-cycle progression by modulating the ARF–MDM2–p53 axis and intersecting with RB/E2F-dependent control of G1/S transcriptional programs. Dmp1 binds promoter elements in genes governing proliferation and checkpoint responses, thereby influencing cellular senescence, stress signaling, and oncogene-induced growth control. Altered DMTF1 activity or dosage has been linked to dysregulated proliferation and tumor suppressor network imbalance, making it a useful node for studying transcriptional control of cell fate decisions in cancer-relevant contexts.
Dmp1 CRISPR/Cas9 KO Plasmid (h) is a pool of plasmids designed for targeted disruption of the DMTF1 gene in human cell lines. Each plasmid co-expresses a unique single guide RNA (sgRNA) targeting a distinct site within the DMTF1 together with the Streptococcus pyogenes Cas9 nuclease. The plasmids also encode GFP, allowing fluorescent identification and enrichment of successfully transfected cells by fluorescence microscopy or flow cytometry.
The multi-guide design increases the likelihood of generating insertions or deletions (indels) that disrupt the DMTF1 open reading frame following Cas9-mediated double-strand break formation. DNA breaks introduced by the CRISPR/Cas9 system are repaired through endogenous non-homologous end joining (NHEJ) pathways, frequently resulting in frameshift mutations that abolish Dmp1 protein expression.
This CRISPR knockout system enables efficient generation of DMTF1-deficient cell models for investigation of Dmp1 signaling, functional genomics studies, cancer biology research, and evaluation of therapeutic responses in human cell lines.
