DMH-1 is a selective inhibitor of ACTR-I, the bone morphogenic protein (BMP) ALK2 receptor. DMH-1 is an analog to the BMP receptor inhibitor, dorsomorphin. Compared to dorsomorphin, DMH-1 is non-selective. DMH-1 has an IC50 = 108 nM and displays no detectable inhibition of AMPK, ALK5, KDR (VEGFR-2) or PDGFRbeta receptors. DMH-1 blocks phosphorylation of Smads 1, 5, and 8 in HEK293 cells induced by BMP4. In zebrafish, DMH-1 was shown to disrupt dorsoventral development during in vitro studies.
