Diphenhydramine hydrochloride blocks the effect of histamine at Histamine H1 Receptor (H1 receptor) sites, which results in an increase in vascular smooth muscle contraction. Diphenhydramine hydrochloride has been shown to have inhibitive tumor promotion properties. The compound decreases peripheral nociceptors sensitization. Diphenhydramine hydrochloride was found to inhibit re-uptake of the neurotransmitter serotonin, which led to a search for viable antidepressants with similar structures and fewer side effects, such as selective serotonin re-uptake inhibitor (SSRI). Diphenhydramine hydrochloride also acts as a sodium channel blocker, which is responsible for its actions as a local anesthetic. The labeled form of this product is also available, Diphenhydramine-d6 Hydrochloride (sc-218244). Diphenhydramine hydrochloride is an inhibitor of mAChR M1.
