Diminutol is a cell-permeable 2,6,9-trisubstituted purine analog that blocks mitotic spindle assembly by competitively inhibiting NQO1, an NADP-dependent oxidoreductase. Diminutol may act to interact with the NADP binding site and directly affect tubulin polymerization. Diminutol does not induce oxidative stress or affect the activity of Cdk1, DNA replication, or actin polymerization. Diminutol may act to cause significant microtubule depolymerization in Xenopus XL177 cells at 50muM.
