BACKGROUND

DAT Antibody (6-8D6) is a rat monoclonal IgG2a antibody that detects the DAT protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-DAT antibody (6-8D6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The dopamine transporter (DAT) plays a crucial role in regulating dopamine levels in the synaptic cleft by facilitating the reuptake of dopamine from the extracellular space back into presynaptic neurons. This function is vital for maintaining dopamine homeostasis, which is essential for normal neurological function and behavior. Dysregulation of DAT activity has been implicated in several neuropsychiatric disorders, including attention deficit hyperactivity disorder (ADHD) and substance use disorders, highlighting DAT′s importance in both health and disease. DAT′s interaction with various neurotoxins underscores its significance in neurodegenerative conditions, particularly in the context of Parkinson′s disease, where DAT dysfunction can lead to the selective vulnerability of dopaminergic neurons.

Genetic locus: SLC6A3 (human) mapping to 5p15.33; Slc6a3 (mouse) mapping to 13 C1.

SOURCE

DAT (6-8D6) is a rat monoclonal antibody raised against amino acids 180-218 corresponding to the second extracellular loop of DAT of human origin.

PRODUCT

Each vial contains 200 µg IgG2a in 1.0 ml of PBS with < 0.1% sodium azide and 0.1% gelatin.

APPLICATIONS

DAT (6-8D6) is recommended for detection of DAT of mouse, rat and human origin by Western Blotting (starting dilution 1:200, dilution range 1:100-1:1000), immunoprecipitation [1-2 µg per 100-500 µg of total protein (1 ml of cell lysate)], immunofluorescence (starting dilution 1:50, dilution range 1:50-1:500) and immunohistochemistry (including paraffin-embedded sections) (starting dilution 1:50, dilution range 1:50-1:500).

Are there reports using this DAT ab for Immunoprecipitation, if so which ones? Could it be used for magnetic isolation by coupling to Dynabeads?

Thank you for your question. DAT Antibody (6-8D6): sc-32259 has been tested and is recommended for IP applications with DAT of mouse, rat and human origin. As of now there are no publications in which this antibody has been used for immunoprecipitation.

Can I use it on rat sections, blocking with normal donkey serum?

Thank you for your question. Yes, this antibody is mouse, rat and human reactive, so using it on rat samples is fine. If you are doing a Western blot we would recommend blocking in 5% milk with TBS-T. You can block in normal donkey serum if you are doing an IHC or IF, especially if you are using a donkey anti-rat secondary antibody. Is a blocking peptide for with this monoclonal antibody available?

Thank you for your question. This DAT antibody sc-32259 is raised against a recombinant protein and not a short synthetic peptide and a blocking peptide is not possible to design for it.
