DAT Antibody (6-8D6) is a rat monoclonal IgG2a antibody that detects the DAT protein of mouse, rat, and human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry. Anti-DAT antibody (6-8D6) is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. The dopamine transporter (DAT) plays a crucial role in regulating dopamine levels in the synaptic cleft by facilitating the reuptake of dopamine from the extracellular space back into presynaptic neurons. This function is vital for maintaining dopamine homeostasis, which is essential for normal neurological function and behavior. Dysregulation of DAT activity has been implicated in several neuropsychiatric disorders, including attention deficit hyperactivity disorder (ADHD) and substance use disorders, highlighting DAT′s importance in both health and disease. DAT (6-8D6) is a rat monoclonal antibody raised against amino acids 180-218 corresponding to the second extracellular loop of DAT of human origin.
Each vial contains 200 µg IgG2a in 1.0 ml of PBS with < 0.1% sodium azide and 0.1% gelatin.

DAT (6-8D6) is recommended for detection of DAT of mouse, rat and human origin by Western Blotting (starting dilution 1:200, dilution range 1:100-1:1000), immunoprecipitation [1-2 µg per 100-500 µg of total protein (1 ml of cell lysate)], immunofluorescence (starting dilution 1:50, dilution range 1:50-1:500) and immunohistochemistry (including paraffin-embedded sections) (starting dilution 1:50, dilution range 1:50-1:500).
