DAN-1 EE hydrochloride is a fluorescent indicator for the bioimaging of intracellular nitric oxide (NO) formation. Upon entry into the cell, DAN-1 EE is transformed by cytosolic esterases into the less cell permeable DAN-1. DAN-1 is a potent, selective agonist of the metabotropic glutamate receptor subtype 5 (mGluR5) and is used to study the effects of mGluR5 activation. The activation of mGluR5 leads to the release of several neurotransmitters, including glutamate, which is involved in the regulation of synaptic plasticity and learning and memory. DAN-1 EE hydrochloride has also been shown to modulate the release of neurotransmitters such as dopamine, serotonin, and acetylcholine, as well as the activity of various ion channels, including calcium and potassium channels, which are involved in regulating neuronal excitability.
