SEE ALSO...

CRY1 Antibody (W-L5) is a mouse monoclonal IgG1 kappa light chain antibody that detects CRY1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry, and enzyme-linked immunosorbent assay (ELISA). CRY1 Antibody (W-L5) is available in a non-conjugated form, specifically designed for researchers seeking to study cryptochrome-1′s role in circadian rhythms. CRY1 functions as a light-independent inhibitor of the circadian clock, interacting with key components such as Clock and BMAL1 to regulate gene expression involved in the light/dark cycle. Located primarily in the nucleus, CRY1 plays a vital role in the negative feedback loop that stabilizes circadian rhythms by forming heterodimers with other proteins like Per1 and Per2, which in turn inhibit Clock-BMAL1 dimer activity. This interaction is essential for proper functioning of the suprachiasmatic nucleus, the brain′s master clock, ensuring that physiological processes such as hormonal rhythms, sleep cycles, and feeding behaviors are synchronized with environmental light cues. Understanding CRY1′s dynamics and interactions is fundamental for elucidating the molecular mechanisms underlying circadian regulation and its implications for health and disease.

Can sc-101006: CRY1 (W-L5) monoclonal antibody be used to stain formalin-fixed, paraffin-embedded (FFPE) tissue sections?

Asked by: Germaine
Thank you for your question. Yes, sc-101006: CRY1 (W-L5) is recommended for use in IHC with paraffin-embedded sections. We recommend performing antigen retrieval with sodium citrate buffer (pH 6) and heat. The full protocol can be found here: https://www.scbt.com/scbt/resources/protocols/immunoperoxidase-staining

I have been having problems producing clean blots with specific bands with this antibody. Is anyone here willing to share their blotting procedure? Thanks in advance!

Asked by: Tyrsett
Thank you for your question. We recommend blocking with Blotto (5% nonfat milk in TBST) for one hour at room temperature, but you can try overnight at 4C if you are experiencing high background. We then recommend diluting the primary antibody in fresh Blotto at a dilution of 1:200 and incubating for one hour at room temperature. We load 50 µg sample protein. And to help reduce non-specific banding, we recommend using our specific mouse IgG kappa binding protein (sc-516102) in place of a conventional anti-mouse IgG secondary antibody. A link to our complete recommended protocol is provided below. https://www.scbt.com/scbt/resources/protocols/western-immuno-blotting If you continue to have trouble, please contact our Technical Service Department and we can assist you with troubleshooting the antibody.
