CNF1 Antibody (NG8) is a mouse monoclonal IgG2a antibody that detects the CNF1 protein of E. coli origin by western blotting (WB). Anti-CNF1 antibody (NG8) is available as the non-conjugated format. Cytotoxic necrotizing factor 1 (CNF1) is a potent toxin produced by uropathogenic and meningitis-causing strains of Escherichia coli, and CNF1′s role in pathogenesis is critical due to its ability to permanently activate Rho-GTP-binding proteins, including Rho, Rac, and Cdc42, through deamidation or transglutamination. This activation leads to significant reorganization of the actin cytoskeleton, resulting in the formation of large stress fibers and increased focal adhesion points, which are essential for cell motility and stability. CNF1 internalization into host cells occurs via endocytosis, followed by transport into late endosomes, where CNF1′s middle domain, characterized by two hydrophobic helices, facilitates translocation across the membrane upon acidification. CNF1′s action extends to potential interference with mitotic processes, contributing to aneuploidy and multinucleation, which are hallmarks of various pathological conditions. CNF1 (NG8) monoclonal antibody enables valuable insights into bacterial pathogenesis mechanisms and cellular responses to this unique toxin.
