The protein i intend to couple to the CNBr-Agarose is diluted in a buffer containing 150 mM of Tris-HCl and NaCl, I understand that Tris-HCl are deleterious for coupling reaction, what can i do to overcome this obstacle and be able to couple my protein?

Thank you for contacting Santa Cruz Biotechnology. To remove the Tris-HCl and NaCl, you might consider a desalting/buffer exchange of your protein into a more suitable buffer. I recommend using a buffer with similar pH to your current so that the protein remains stable. We have multiple prepacked Desalting columns that can accommodate different sample volumes. I have listed some of those options below. Whether it will work for your sample will partly depend on the size of your target protein, and will require testing. HiTrap Desalting columns with Sephadex G-25 resin - Offered in 5mL bed volume, these are easy to use with syringe or chromatography system. It can take samples up to ~1.5mL HiPrep Desalting columns with Sephadex G-25 resin - This is a larger ~53mL column that can take samples up to ~15mL.

Can I immunize rabbits using CNBr activated sepharoes + any protein?Can I replace Freund adjuvans by this way? Thank you and regards JD

Thank you for your question. We would not recommend using these beads for immunization applications. CNBr-Activated Agarose is intended to use for conjugating antibodies to agarose beads for immunoprecipitation research applications.

I want to know the reaction between CNBr-bead and a chemical substance without OH-group and NH2-group. I want to know whether reaction between CNBr-bead and CH3-group can occur.
