Chlorantraniliprole is a unique insecticide classified as an anthranilic diamide. It acts as an agonist of ryanodine receptors, which are present in the sarcoplasmic reticulum of muscle cells and endoplasmic reticulum of non-muscle cells. Unlike ryanodine, it binds to a distinct site on the receptor and stimulates the release of calcium from intracellular stores. This compound demonstrates a high selectivity for insect ryanodine receptors compared to mammalian ones. Its efficacy is evident with EC50 values ranging from 40 to 50 nM for P. americana neurons and H. virescens or D. melanogaster recombinant ryanodine receptors, whereas the EC50s for C2C12 mouse, PC12 rat, and IMR32 human cells are 14,000 nM, >100 µM, and >100 µM, respectively.
