CD73 (IE9) is a mouse monoclonal IgG3 kappa light chain antibody that detects CD73 of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and immunohistochemistry with paraffin-embedded sections (IHCP). Anti-CD73 antibody (IE9) is available in non-conjugated form. CD73 (also designated 5′-nucleotidase, E5NT, NT, NT5, NTE, eN and eNT) is a glycosyl-phosphatidylinositol (GPI)-anchored adhesion protein that catalyzes the dephosphorylation of extracellular purine and pyrimidine nucleotides to their corresponding bioactive nucleosides. CD73 exists as a dimer of two identical subunits that depends on GPI to link with the external face of the plasma membrane. Similar to other GPI-anchored proteins, CD73 mediates co-stimulatory signals in T cell activation. CD73 has few structural variants, yet elicits diverse biological function through differential regulation in endothelial cells (EC), subpopulations of B and T cells, germinal center follicular dendritic cells and on thymic medullary reticular fibroblasts. IgG mediated neutralization of CD73 interferes with lymphocyte adhesion to EC, and blocks aggregation of germinal center B cells and follicular dendritic cells. Additionally, IgG-mediated targeting of lymphocyte CD73, but not of endothelial cell CD73, causes shedding of CD73 and tyrosine phosphorylation of proteins.

Est-ce que Anticorps CD73 (IE9): sc-32299 peut être utilisé pour des tests ELISA ?  En quelle quantité est-t-il fourni ? 1ml ?

Merci pour votre question. Anticorps CD73 (IE9): sc-32299 est recommandé pour la détection de CD73 d'origine human par WB, IP, IF et IHC(P); non cross-reactif avec mouse CD73. Nous n'avons pas de données pour soutenir l'utilisation dans ELISA. L'anticorps est fourni à 200 µg dans 1 ml.

I used CD73 Antibody (IE9): sc-32299 for my research. I used auto IHC machine. It is of Roche Company. As I know, the antibody will help to stain cell membrane and sometimes cytoplasm. However, my samples showed nuclear staining. Give me some advices!

Hello: Was CD73 Antibody (IE9): sc-32299 tested on rat tissues? It was found to inhibit the catalytic activity of CD73 (in http://www.jimmunol.org/content/191/8/4165.long). This would be great if I could use it in rat tissue homogenates? thanks, Fran.

Thank you for your question. We do not have any data indicating that this mouse monoclonal antibody, CD73 (IE9): sc-32299, is reactive with rat CD73. We recommend the antibody for detection of CD73 of human origin only. In the publication you have cited, the authors specifically mention that IE9 clone "failed to detect the murine CD73 variant ".
