CD4 Antibody (GK1.5) is a rat monoclonal IgG2b antibody designed to recognize the extracellular region of the CD4 protein, also known as T4 or Leu 3, across mouse, rat, and human species. Raised against CTL clone V4 cells of mouse origin, anti-CD4 antibody (GK1.5) is recommended for use in immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM), ensuring precise detection and analysis of CD4 in various experimental settings. CD4 is a membrane-bound glycoprotein composed of four extracellular immunoglobulin-like domains, playing a crucial role in the immune system by acting as a co-receptor for the T cell receptor (TCR). This interaction enhances the binding affinity between TCRs and class II major histocompatibility complex (MHC) molecules on antigen-presenting cells, which is essential for effective T cell activation and the subsequent immune response. CD4 interacts with several key proteins, including the tyrosine kinase Lck, which associates with its cytoplasmic tail to initiate downstream signaling pathways necessary for T cell activation. Additionally, CD4 forms complexes with other co-receptors such as CD3 and CD28, facilitating signal transduction and T cell co-stimulation. The ability of CD4 to interact with these proteins underscores its importance in both adaptive immunity and the pathogenesis of diseases like HIV, where CD4 serves as a primary receptor for viral entry. Beyond T helper cells, CD4 is also expressed on cortical cells, mature medullary thymocytes, microglial cells, and dendritic cells, highlighting its broad role in immune regulation.
