CD17 Antibody (MEM-74) is a mouse monoclonal IgM antibody that detects CD17 protein of human origin by flow cytometry (FCM). CD17, also known as Lactosylceramide or LacCer, plays a crucial role as an intermediate glycosphingolipid in the metabolism of higher gangliosides, and is primarily localized to sphingolipid-sterol rafts within the cell membrane. Such localization facilitates the clustering of signaling molecules and receptors, essential for various cellular processes, including cell adhesion, migration, and communication. CD17 is expressed in several immune cell types, including monocytes, granulocytes, basophils, platelets, and a subset of peripheral B cells (CD19+), as well as tonsillar dendritic cells. CD17 expression rapidly downregulates on activated granulocytes while upregulating on interleukin-2 activated T lymphocytes, indicating a dynamic role in immune responses. CD17 binds to bacteria, suggesting involvement in phagocytosis, and can be produced by vascular endothelial cells in response to vascular endothelial growth factor (VEGF), mediating signaling pathways that promote PECAM-1 expression and angiogenesis. In neuroinflammation, tumor necrosis factor alpha (TNFα) can induce astrogliosis, leading to increased intracellular levels of CD17, which may contribute to spinal cord injury pathophysiology. Aberrant glycosphingolipid levels, including CD17, are often associated with cancer cells, potentially influencing integrin clustering and internalization, thereby impacting tumor progression and metastasis. CD17 (MEM-74) antibody serves as an invaluable tool for researchers studying these complex biological processes.
