CD15 Antibody (HI98) is a mouse monoclonal IgM kappa light chain antibody that detects CD15 in mouse, rat, and human samples through various applications including western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and flow cytometry (FCM). CD15, also known as Lewis X or LeX, is a carbohydrate antigen that plays a crucial role in cell adhesion and interaction, primarily found on the surface of leukocytes and certain tumor cells. This trisaccharide is synthesized by the action of fucosyltransferase IV (FucT-IV), which transfers an α-fucose residue onto the β-GlcNAc moiety of cellular N-acetyllactosamines. The presence of CD15 on cell surfaces is essential for mediating calcium-dependent homotypic binding, allowing cells with high CD15 expression to aggregate effectively in the presence of calcium ions. Furthermore, CD15 serves as a ligand for selectins, which are critical in facilitating leukocyte-specific interactions during immune responses. The ability of CD15 to promote cell adhesion and aggregation underscores its importance in various physiological processes, including inflammation and immune cell trafficking. Anti-CD15 antibody (HI98) is available in both conjugated and non-conjugated forms, providing flexibility for different experimental needs.

Thank you for your question. You can find our recommended protocols by following this link: https://www.scbt.com/scbt/resources/protocols?&_requestid=1574639. When working with formalin-fixed and/or paraffin-embedded tissue samples we recommend antigen retrieval using heat treatment with 10 mM sodium citrate buffer, pH 6.0 (sc-294091); or with 50 mM glycine-HCl buffer (sc-29096), pH 3.5, with 0.01% (w/v) EDTA (sc-29092).

Can CD15 (HI98): sc-19649 monoclonal antibody be used in IF or IHC with mouse tissue or cells? Will there be non-specific staining?

Thank you for your inquiry. The use of mouse monoclonal antibodies with mouse samples is very common and typically poses no problem. To eliminate any potential cross-reactivity of an anti-mouse secondary detection reagent, CD15 (HI98): sc-19649 is available in a variety of direct conjugations, such as HRP, PE, FITC and AlexaFluors.
