CAY10500 is an inhibitor of tumor necrosis factor α (TNFα) that hinders its binding to TNF Receptor 1 (TNFR1). TNFα acts as a central mediator in various inflammatory conditions, including rheumatoid arthritis, toxic shock, and sepsis. Functioning as a trimer, TNFα facilitates receptor trimerization, which triggers proinflammatory and/or apoptotic signaling pathways. By binding to the biologically active TNFα trimer, CAY10500 accelerates the displacement of a single subunit, leading to the rapid inactivation of the cytokine. In a cell-based assay, CAY10500 effectively inhibited TNFα-induced stimulation of IKB degradation. Notably, both TNFα and the angiotensin II/angiotensin II receptor type 1 (AT1) axis play roles in neuropathic pain and nociception.
