Calponin 1/2/3 Antibody (G-10) is a mouse monoclonal IgG1 kappa light chain antibody that detects Calponin 1 protein of mouse, rat, and human origin, as well as Calponin 2 and Calponin 3 proteins of human origin by western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), immunohistochemistry with paraffin-embedded sections (IHCP), and enzyme-linked immunosorbent assay (ELISA). Calponin 1/2/3 (G-10) antibody is available in both non-conjugated and various conjugated forms, including agarose, horseradish peroxidase (HRP), phycoerythrin (PE), fluorescein isothiocyanate (FITC), and multiple Alexa Fluor® conjugates. Calponin plays a crucial role in regulating smooth muscle contraction and serves as a marker for smooth muscle cell differentiation. Calponin is primarily located in smooth muscle tissue, where Calponin interacts with actin and tropomyosin, functioning as an inhibitory factor for smooth muscle actomyosin activity. This interaction is vital for maintaining the contractile state of smooth muscle cells, as Calponin modulates the activity of phosphorylated myosin by inhibiting the actin-activated MgATPase activity. Notably, Calponin′s regulatory properties are compromised upon phosphorylation, particularly at Serine 175, by protein kinase C or calmodulin-dependent protein kinase II, which can lead to altered smooth muscle function. The three isoforms of Calponin—Calponin 1 (basic), Calponin 2 (neutral), and Calponin 3 (acidic)—are distributed in various tissues, with Calponin 2 also found in heart muscle and Calponin 3 in the brain, highlighting the diverse roles in different physiological contexts.
