CalciFluor™ Rhod-4, AM is a fluorescent calcium indicator developed as an improvement to Rhod-2 AM. Accurate measurement of calcium is vital for a wide range of biological investigations. To enable the study of intracellular free calcium concentrations, fluorescent probes with spectral responses upon binding Ca2+ have been instrumental. Techniques such as fluorescence microscopy, flow cytometry, fluorescence spectroscopy, and fluorescence microplate readers have utilized these probes. Among the red fluorescent calcium indicators, Rhod-2 is employed however, its live cell fluorescence and cellular calcium responses are modest. To address these limitations, Rhod-4™ has been developed, maintaining the spectral wavelength of Rhod-2 while enhancing cell loading and calcium response. In both CHO and HEK cells, Rhod-4™ AM exhibits a cellular calcium response that is ten times more sensitive than Rhod-2 AM, making it useful for calcium imaging studies.
