


Cabin-1 Antibody (E-11) is a mouse monoclonal IgG1 antibody that detects Cabin-1 in mouse, rat, and human samples through applications such as western blotting (WB), immunoprecipitation (IP), immunofluorescence (IF), and enzyme-linked immunosorbent assay (ELISA). Cabin-1, also known as calcineurin binding protein, plays a crucial role in regulating calcineurin activity, a serine/threonine phosphatase essential for T cell signaling and apoptosis. Cabin-1 is primarily located in the nucleus, where Cabin-1 interacts with calcineurin to modulate activity in response to changes in intracellular calcium levels and protein kinase C activity. This regulation is vital because calcineurin is responsible for transcriptional activation of key cytokines and transcription factors, including NFAT, NFkB, and AP-1, which are integral to T cell receptor-mediated signaling. When T cell receptor activation occurs, protein kinase C induces Cabin-1 hyperphosphorylation, enhancing binding affinity to calcineurin. This binding forms a complex that inhibits calcineurin′s activity, thereby attenuating T cell receptor-mediated signaling. Additionally, Cabin-1 directly associates with MEF-2 proteins, a family of transcription factors critical for regulating apoptosis signaling in T cells. The interaction between Cabin-1 and MEF-2 not only inhibits MEF-2-mediated gene transcription but also plays a significant role in preventing apoptosis, highlighting Cabin-1′s importance in maintaining T cell homeostasis and function.
