c-Fos Antibody (554C1a) is a mouse monoclonal IgG1 antibody raised against the C-terminal region of human c-Fos, a nuclear phosphoprotein crucial for transcriptional regulation. Anti-c-Fos antibody (554C1a) demonstrates specificity for human c-Fos in Western blotting (WB) and immunoprecipitation (IP) applications. As an immediate early gene product, c-Fos responds rapidly but transiently to various stimuli including growth factors, cytokines, and stress signals. Unlike c-Jun which can form both homo- and heterodimers, c-Fos functions exclusively through heterodimerization with Jun family members to form the Activator Protein-1 (AP-1) transcription factor complex. The sophistication of c-Fos regulation extends through interactions with functional homologs Fra-1, Fra-2, and FosB, as well as select ATF and CREB family members, forming leucine zipper dimers with varied affinities for AP-1 and CRE DNA binding sites. This diversity in complex formation enables precise control of gene expression programs governing cell proliferation, differentiation, and survival. c-Fos maintains strict nuclear localization, essential for c-Fos′s role in driving cell cycle progression and differentiation. When c-Fos regulation becomes disrupted, particularly through overexpression, the resulting dysregulation can contribute to oncogenesis. Through these diverse cellular functions, c-Fos (554C1a) antibody serves as a valuable tool for investigating transcriptional regulation and cancer biology.
