Bumetanide (Ro 10-6338) is a specific inhibitor of Na+-K+-2Cl- cotransporter NKCC1, which may act to bind with high affinity and saturate binding of the cotransporter. The inhibitor has been reported to bind to different cell membranes, including HeLa cells, vascular muscle cells, endothelial cell, erythrocyte cells, and colonic adenocarcinoma cells, among others. Furthermore, Bumetanide has been observed to block the depolarization of GABA in cortical development studies.
